Polymorphic nanobody crystals as long‐acting intravitreal therapy for wet age‐related macular degeneration

Wet age-related macular degeneration (wet AMD) is the most common cause of blindness, and chronic intravitreal injection anti-vascular endothelial growth factor (VEGF) proteins has been dominant therapeutic approach. Less a prolonged inter-injection interval are main drivers behind new wet AMD drug innovations. By rationally engineering surface residues model anti-VEGF nanobody, we obtained series nanobodies with identical protein structures VEGF binding affinities, while drastically different crystallization propensities crystal lattice structures. Among these nanobody crystals, P212121 appeared to be denser released slower than P1 lattice, crystals embedding zinc coordination further slowed release rate. The polymorphic could potentially breakthrough strategy for administration proteins.
